Foundayo™ (orforglipron) is the first non-peptide oral GLP-1 receptor agonist, FDA-approved in 2026 for chronic weight management. In the 72-week ATTAIN-1 clinical trial, adults without diabetes taking the highest 17.2 mg dose of Foundayo lost an average of 11.1% of body weight (about 25 lbs at average starting weight). Yes. Foundayo may reduce the effectiveness of oral birth control pills. The manufacturer recommends using an additional non-hormonal method of contraception (such as condoms) or switching to a non-oral form of birth control for 30 days after starting Foundayo and for 30 days after each dose increase. The most common side effects are nausea, constipation, diarrhea, vomiting, indigestion, abdominal pain, headache, bloating, fatigue, belching, heartburn, gas, and hair loss. GI side effects are most pronounced during dose increases and typically improve once you stabilize at a given dose. Slow titration significantly reduces severity. Persistent or severe symptoms should always be reported to your provider.
Foundayo shares the same GLP-1 class contraindications: personal or family history of medullary thyroid carcinoma (MTC), Multiple Endocrine Neoplasia type 2 (MEN2), and pregnancy. It should be used with caution in patients with a history of pancreatitis, severe renal impairment, or active gallbladder disease. Foundayo should not be combined with other GLP-1 medications. Do not use if you have a personal or family history of medullary thyroid carcinoma (MTC) or Multiple Endocrine Neoplasia syndrome type 2 (MEN 2). Common side effects include nausea, constipation, diarrhea, vomiting, indigestion, and abdominal pain. Pancreatitis, gallbladder disease, dehydration, and acute kidney injury have been reported.
